Description: Transgenic line expressing SOMAT679A, the phosphosite mutant form of SOMA (sensor of MAPK activity), with a nuclear exclusion signal. For targeted expression of the sensor in the cytoplasm of transgenic plants, the SOMAT679A protein coding sequence was moved into a derivative of the binary vector pRI 201-AN (Takara Bio, USA) to produce pSOMAT679A-NES. Col-0 plants were transformed by the floral dip method and the transgenic line was identified by screening T1 seed for seedlings showing YFP fluorescence. Genotype of the transgene is unknown.

Growth requirement: This T-DNA line expresses a FRET sensor for monitoring MAP kinase activity in Arabidopsis. Because gene silencing can affect the expression of the FRET sensor, one should always screen young seedlings on agar plates for YFP or CFP fluorescence to identify individual seedlings in which the sensor is being properly expressed before doing an imaging experiment. When bulking seeds for these lines, one should also screen the individual seedlings used to bulk up seed for YFP or CFP fluorescence.
